Any advice on setting the lighting from the the background reflector for graduated light on the backdrop?

Should it be underexposed by a stop, over or balanced 1:1 with the main light?

depend what you want and how you want it.

eg. light on blue background

under one stop = dark blue
same as main light = blue
over one stop = light blue

so you decide where you want to place your light to get the graduated effect

Another question. Where to take the meter reading from for the background? Incident or reflected?

My guess is incident (dome facing the camera) with the lightmeter against the backdrop.
