If I am shooting something where I will be taking a photo or two every 5min-20mins interval, should I switch off my camera or leave it to auto power off? My auto power-off is set to 1min. Will leaving it in the auto off mode drain a lot more battery?

Well, I'd say leave it to auto power off, because you won't want to miss the moment when it comes!

Timed sleep mode will definitely drain power during active period but does not cause problem in reality unless it active time is too long. But if you camera is always subjected to bumps, the constant button pressing may not put it to sleep.
Products are tuned for optimal usage pattern, so you will know the product power draining pattern as you use with time.

For me, auto power off is not an issue.

i did not turn off my camera for an entire week when i dumped it into the dry cabi. meaning it was in standby mode all the while.

it went in after 500++ shots, and had 3/4 battery.

after i took it out the next weekend, it was still displaying 3/4 batt.

waste battery or not, its up to you how you define it. cos to me, its not.
